The present invention relates to a pharmaceutical composition and health-aid food comprising organic solvent fractions of the underground parts of Platycodon grandiflorum or red Platycodon grandiflorum as an active ingredient for preventing or treating thrombosis. More particularly, the present invention relates to a pharmaceutical composition and health-aid food comprising organic solvent fractions of Platycodon grandiflorum or red Platycodon grandiflorum, obtained by forming an ethanol extract of Platycodon grandiflorum or red Platycodon grandiflorum prepared by repeatedly digesting and drying Platycodon grandiflorum, and carrying out sequential fractionation thereof with an organic solvent, as an active ingredient for preventing or treating/improving thrombosis. The organic solvent fractions of the underground parts of Platycodon grandiflorum or red Platycodon grandiflorum as an active ingredient for preventing or treating thrombosis shows strong anti-thrombotic activity derived from the effects of inhibiting thrombus formation-related enzymes and inhibiting agglomeration of blood platelets functioning as an initiator of thrombus formation, as shown by examples. In addition, the organic solvent fractions improve blood flow and can be useful for preventing and treating thrombosis such as ischemic stroke and hemorrhagic stroke. In addition, the organic solvent fractions have high thermal stability and causes no loss of anti-thrombotic activity even under an acidic condition of pH 2 and in plasma, and thus can be processed and formulated into various shapes, such as an extract, powder, pill and tablet, to be taken frequently.
