The present invention relates to methods, compositions and kits that are associated with resistance to treatment with anti-cancer agents, particularly BRAF inhibitors. In particular embodiments, this invention relates to a mutation in a BRAF sequence that confers resistance to the BRAF inhibitor. Identification of these mutations in the BRAF sequence allows identification and planning of next generation BRAF inhibitors.
