A method of preparing a lactase preparation, comprising a. using mutagenesis conditions or recombinant gene manipulation techniques in a culture that produces arylsulfatase, so that part of the culture is modified to form an arylsulfatase deficient host cell, in which the progenitor host cell comprises one or more nucleotide sequences that encode arylsulfatase , and the mutant host cell produces less arylsulfatase activity than the parent cell when cultured under the same conditions； and isolating the host cell； b. cultivating said host cell in a nutrient medium, under conditions conducive to lactase expression, and expressing lactase in said host cell； and c. recover lactase from the nutrient medium or from the host cell.Un procedimiento para preparar una preparación de lactasa, que comprende a. usar condiciones de mutagénesis o técnicas de manipulación genética recombinante en un cultivo que produce arilsulfatasa, de manera que parte del cultivo se modifica para formar una célula hospedante deficiente en arilsulfatasa, en el que la célula hospedante progenitora comprende una o más secuencias nucleotídicas que codifican arilsulfatasa, y la célula hospedante mutante produce menos actividad de arilsulfatasa que la célula progenitora cuando se cultivan en las mismas condiciones； y aislar la célula hospedante； b. cultivar dicha célula hospedante en un medio nutriente, en condiciones que conduzcan a la expresión de la lactasa, y expresar la lactasa en dicha célula hospedante； y c. recuperar la lactasa del medio nutriente o de la célula hospedante.
