The present invention provides novel multispecific antibody platforms, multispecific antibodies and methods for preparing the antibodies. The CH3 domain containing the polypeptide chain of the Fc region is modified by increasing the positive charge of one chain and the negative charge of another. The formation of a heterodimer in co-transfection with this modified chain corresponds to the formation of a wild-type antibody when transfected into the same cell line with a wild-type polypeptide. Furthermore, the modification provides a simple one-step purification for heterodimers.本発明は、新規な多重特異性抗体プラットフォーム、多重特異性抗体およびその抗体を調製する方法を提供する。１つの鎖の正電荷および別の１つの鎖の負電荷を増加させることにより、Ｆｃ領域のポリペプチド鎖を含むＣＨ３ドメインを修飾する。この修飾された鎖を用いたコトランスフェクションにおけるヘテロ二量体の形成は、野生型ポリペプチドを用いて同じ細胞系にトランスフェクトされた場合の野生型抗体の形成に相当する。さらに、前記の修飾はヘテロ二量体に対して簡単な一段階精製を提供する。
