The present invention relates to substituted azole derivatives in combination with cytokines in the ex vivo expansion of CD34+ hematopoietic stem and progenitor cells (HSPC) in a biological sample, more particularly the expansion of these cells obtained from non-enriched, i.e., the mononuclear fraction of the biological sample. The present invention further describes the transplantation regimen of the expanded hematopoietic graft developed through xenotransplantation studies. In a preferred embodiment, the combination comprising the azole based compounds and cytokines selected from SCF, TPO, FLT-3L and IGFBP-2 and results in the expansion of expansion of CD45+CD34+CD38-CD45RA-CD90+ hematopoietic stem cells and/or CD45+CD34+CD38-CD45RA-CD90+CD49f+ hematopoietic stem cells and/or CD45+CD34+CD38-CD45RA- hematopoietic progenitor cells from the mononucleated cells isolated from umbilical cord blood.La présente invention concerne des dérivés d'azole substitués en combinaison avec des cytokines pour l'expansion ex vivo de cellules souches et progénitrices hématopoïétiques CD34+ dans un échantillon biologique, plus particulièrement l'expansion de ces cellules obtenues à partir d'une fraction non enrichie, c'est-à-dire la fraction mononucléaire de l'échantillon biologique. La présente invention concerne en outre le régime de transplantation du greffon hématopoïétique expansé développé par des études de xénogreffe. Dans un mode de réalisation préféré, la combinaison comprend les composés à base d'azole et les cytokines choisies parmi SCF, TPO, FLT-3L et IGFBP-2 et conduit à l'expansion de cellules souches hématopoïétiques CD45+CD34+CD38-CD45RA-CD90+ et/ou de cellules souches hématopoïétiques CD45+CD34+CD38-CD45RA-CD90+CD49f+ et/ou de cellules progénitrices hématopoïétiques CD45+CD34+CD38-CD45RA- issues des cellules mononucléées isolées à partir de sang de cordon ombilical.
