This invention relates to methods of treating cancer in a subject in need thereof, e.g., in a human in need thereof, comprising determining at least one of the following in a sample from said human: (a) the presence or absence of a mutation at the alanine 677 (A677) residue in EZH2 in a sample from said human or (b) the presence or absence of a mutation at the tyrosine 641 (Y641) residue in EZH2 or (c) the presence or absence of an increased level of H3K27me3 as compared to a control, and administering to said human an effective amount of an EZH2 inhibitor or a pharmaceutically acceptable salt thereof if at least one of said A677 mutation, Y641 mutation, or increased level of H3K27me3 is present in said sample.Cette invention concerne des méthodes de traitement du cancer chez un sujet en ayant besoin, par exemple chez un être humain en ayant besoin, comprenant la détermination dau moins un des éléments suivants dans un échantillon provenant dudit être humain : (a) la présence ou labsence dune mutation au niveau du résidu alanine 677 (A677) dans EZH2 dans un échantillon provenant dudit être humain ou (b) la présence ou labsence dune mutation au niveau du résidu tyrosine 641 (Y641) dans EZH2 ou (c) la présence ou labsence dun niveau accru dH3K27me3 en comparaison à un témoin, et ladministration audit être humain dune quantité efficace dun inhibiteur dEZH2 ou dun sel pharmaceutiquement acceptable de celui-ci si au moins un de ladite mutation A677, ladite mutation Y641 ou le niveau accru dH3K27me3 est présent dans ledit échantillon.
