The present invention relates to a method for the proliferation of regulatory T cells (Treg) in vitro by an autologous antigen-presenting cell (APC), and uses thereof. More specifically, the method, which is used for the in-vitro proliferation of the CD4^+CD25^highCD127^- Treg, includes the following steps (a) preparing the CD4^+CD25^highCD127^- Treg and the APC separately and (b) stimulating the CD4^+CD25^highCD127^- Treg with the APC, anti-CD3 antibody, anti-CD28 antibody, interleukin2 (IL2), and TGF-beta for proliferating the CD4^+CD25^highCD127^- Treg. The present invention also relates to an immunosuppressive composition and a pharmaceutical composition for the prevention or treatment of autoimmune diseases or inflammatory diseases.
