PURPOSE: A preservation method of cryptomeria japonica seed at super low temperature is provided to facilitate sprouting and to improve germination rate value. CONSTITUTION: A preservation method of cryptomeria japonica seed at super low temperature comprises: a step of dipping cryptomeria japonica seed in super low temperature cryoprotective agent solution and storing the cryptomeria japonica seed in liquid nitrogen； a step of thawing the cryptomeria japonica seed stored in the liquid nitrogen； and a step dipping the thawed cryptomeria japonica seed in polyethylene glycol solution.본 발명은 삼나무종자(Japanese cedar seed)의 초저온 저장방법에 관한 것으로 보다 상세하게는 삼나무종자를 초저온 동결보호제 용액에 침지시킨 후 액체질소에 저장하는 단계； 상기의 액체질소에 저장한 나무종자를 해동시키는 단계； 및 상기의 해동한 삼나무종자를 폴리에틸렌글리콜(PEG) 용액에 침지하는 단계를 포함하는 것을 특징으로 하는 삼나무종자의 초저온 저장방법에 관한 것이다.본 발명은 초저온 저장기술을 이용하여 삼나무종자를 저장함에 있어서 초저온 동결보호제 및 해동한 삼나무종자를 폴리에틸렌글리콜 용액으로 프라이밍(priming) 처리를 이용하여 초저온이 아닌 보통의 방법으로 저장한 삼나무종자와 대비시 동등 수준 이상의 발아율을 가지고, 발아가 빨리 진행되며, 발아율 값(Germination value) 특성이 우수한 삼나무종자의 초저온 저장방법을 제공할 수 있다.
