PROBLEM TO BE SOLVED: To provide a technique of generating a further uniform cell cluster of differentiation from pluripotent cells derived from humans.SOLUTION: The invention comprises the following: a technique for generating populations of differentiated neurons, glial cells, and astrocytes from neuronal precursor cells by initiating differentiation in a stem cell population (embryonic stem cells, etc.) and culturing in a mixed solution under growth conditions for establishing a neuronal precursor cell population; a technique for further differentiating precursor cells by culturing into a variety of different neuronal phenotypes including dopaminergic neurons, and a technique for mass producing cell clusters of differentiation or neuronal precursor cells for drug screening and for the treatment of neurological diseases; a cell cluster acquired at a stage where primate pluripotent stem (pPS) cells are differentiated in a culture medium including two or more ligands which bind to a growth factor receptor selected from antibodies against EGF, bEGF, PDGF, IGF-1 and receptors of ligands thereof; and a method of culturing the cell cluster in a medium containing a cAMP activating substance, a neurotrophic factor or a combination thereof.SELECTED DRAWING: None【課題】ヒト由来の多能性細胞から、より均一な分化細胞集団を生じさせる技術の提供。【解決手段】幹細胞集団(胚性幹細胞など)を、分化を惹起し、神経前駆細胞集団を確立する成長条件の混液中での培養によって、神経前駆細胞、分化したニューロン、グリア細胞及びアストロサイトの集団を生じさせる技術。前駆細胞を培養下で、ドーパミン作動性ニューロンを含む様々な異なる神経表現型へと更に分化させる技術及び分化細胞集団又は神経前駆細胞を、薬物スクリーニング及び神経疾患の治療に用いる為に大量に作製できる技術。EGF、bEGF、PDGF、IGF-1及びこれらのリガンドの受容体に対する抗体から選択する、成長因子受容体と結合する2つ以上のリガンドを含む培地霊長類多能性幹(pPS)細胞を分化させる段階によって得る前記細胞集団。更に前記細胞集団をcAMP活性化物質、神経栄養因子又はそれらの組み合せを含む培地中で培養する方法。【選択図】なし
