Discloses nanobody polypeptides that specifically bind the human CXCR4. The nanobodies are generated by a method, comprising at least the steps of: 1. immunizing a Llama with whole cells that are alive and express the humanCXCR4 on their surface; 2. selecting for binding for human CXCR4; and where 3. the polypeptide displaces more than 50%, of human CXCL12 at high concentrations (? 100 nM) when bound to human CXCR4 in a competition binding assay; and/or 4. the polypeptide fully antagonizes the chemoattractant effects of human CXCL12 to human CXCR4 in a concentration that is equal or lower than 100 nM; and 5. where if the polypeptide comprises two or more nanobody, the polypeptide further comprises one or more peptide linkers Further discloses the nanobody framework and CDR regions comprising SEQ IDs as disclosed. Further discloses are pharmaceutical compositions of the nanobodies and a use of the nanobody in the manufacture of a medicament to treat cancer or AIDS. A nanobody is an antibody fragment consisting of a single variable antibody domain.
