The present invention relates to an anti-CD19 antibody having a variant Fc region having some specific amino acid modifications relative to a wild-type Fc region which confer one or several useful effector functions. The present invention relates in particular to chimeric, humanized or full human anti-CD19 antibodies comprising such a variant Fc region. It relates advantageously to antibodies with an interesting and valuable glycosylation profile, especially a low fucose level and/or a high oligomannose level and low level of sialic acid, high ADCC function and no CDC. The present invention also relates to the use of these antibodies in the treatment, prevention or management of diseases or disorders, such as cancer, especially a B-cell malignancy and auto-immune disease.La présente invention concerne un anticorps anti-CD19 comportant une variante de région Fc qui présente certaines modifications spécifiques dacides aminés par rapport à une région Fc de type sauvage, lui conférant une ou plusieurs fonctions effectrices utiles. La présente invention concerne en particulier des anticorps anti-CD19 chimériques, humanisés ou entièrement humains comprenant une telle variante de région Fc. Elle concerne de façon avantageuse des anticorps présentant un profil de glycosylation utile et intéressant, spécialement un faible taux de fucose et/ou un taux élevé doligomannose et un faible taux dacide sialique, une forte fonctionnalité ADCC et aucune fonctionnalité CDC. La présente invention concerne également lemploi de ces anticorps dans le traitement prophylactique ou thérapeutique ou la gestion de maladies ou de troubles tels que le cancer, spécialement une tumeur maligne des cellules B, et les maladies auto-immunes.
