Disclosed are a controlled release gene delivery composition produced by solubilizing hydrophilic nucleic acid genes in an organic solvent and encapsulating the same in hydrophobic particles and a method for producing the same. The controlled release gene delivery composition according to an embodiment of the present invention comprises: a cationic and hydrophilic polymer grafted with hydrophobic molecules; and a gene composite which is produced by the bonding of genes on an organic solvent mixture. The cationic and hydrophilic polymer and gene composite are dispersed on the organic solvent so as to be easily encapsulated in nano particles.친수성 핵산 유전자를 유기용매에 가용화시키고, 이를 소수성 미립자에 봉입한 제어방출형 유전자 전달용 복합체와 이의 제조방법이 개시된다. 본 발명의 일 실시예에 따른 유전자 전달용 복합체는 소수성 분자로 그래프트된 양이온성이며 친수성 고분자; 및 유전자가 유기용매 혼합물 상에서 결합되어 형성되는 유전자 복합체가 유기용매 상에 분산되어 나노입자 내로 용이하게 봉입되는 것을 특징으로 한다.
