The present invention provides a method of predicting a clinical response fortreatment of acancer with an alkylating chemotherapeutic agent. The method comprisesdetermining the stateof methylation of the O6-methylguanine-DNA methyltransferase (MGMT) gene inthe cancer,wherein methylation of the MGMT gene is predictive of an effective or apositive clinicalresponse to the alkylating chemotherapeutic agent in the treatment of thecancer. Figure 1 showsthe overall survival among subjects with gliomas treated with carmustine, andthe time ofprogression of disease according to the methylation status of the MGMTpromoter.
