The invention is in the field of functional screening of protective antigens against pathogens in particular against pathogens such as parasites, bacteria, viruses or protective antigens of cancer cells in order to identify antigens suitable for the elicitation of a protective immune response in a host, and/or to optimize antigen design, in particular to define suitable antigen combinations for a protective immune response. The invention thus involves using a lentiviral vaccine platform for induction of a specific cellular or humoral response against assayed antigens on one hand, and using a fast and reproducible assay for the evaluation of the induced protective effect after challenge.L'invention concerne le domaine du criblage fonctionnel d'antigènes protecteurs contre des pathogènes, en particulier contre des pathogènes tels que des parasites, des bactéries, des virus, ou d'antigènes protecteurs contre des cellules cancéreuses, dans le but d'identifier des antigènes capables d'induire une réponse immunitaire protectrice chez un hôte et/ou d'optimiser la conception d'antigènes, en particulier pour définir des associations d'antigènes permettant d'obtenir une réponse immunitaire protectrice. L'invention entraîne ainsi l'utilisation d'une plate-forme vaccinale de vecteurs lentiviraux permettant l'induction d'une réponse cellulaire ou humorale spécifique contre des antigènes testés d'une part, et l'utilisation d'un test rapide et reproductible pour l'évaluation de l'effet protecteur induit après la mise en contact avec l'antigène.
