The present invention relates to a cancer microenvironment-sensitive complex drug delivery system containing a novel lipid-peptide-polymer conjugate as a substrate of a cancer overexpression enzyme. The drug delivery system provided according to an aspect of the present invention, compared to an existing drug delivery system in which a ligand targeting a receptor of disease cells is bound to a distal end of a drug carrier, has a large amount of carriers that reaches a target cell and drugs that penetrates into a cell after reaching the target cell, thereby having an effect of improving cancer treatment efficiency.암 과발현 효소의 기질로서 신규 지질-펩티드-고분자 컨쥬게이트가 함유된 암 미세환경 감응성 복합 약물전달시스템에 관한 것으로, 본 발명의 일 측면에 따라 제공되는 약물전달시스템은, 질병 세포의 수용체를 표적하는 리간드가 약물전달체의 말단부에 결합된 종래의 약물전달시스템에 비해, 상대적으로 표적 세포에 도달하는 전달체의 양과, 표적 세포에 도달한 후 세포 내로 투과되는 약물의 양이 많아 암 치료 효율이 개선되는 효과가 있다.
