PROBLEM TO BE SOLVED: To provide methods of reducing levels of a titratable selectable pressure required, the number of amplification cycles, and the time taken to produce desired protein-expressing cell lines.SOLUTION: Codon adaptation of desired protein open reading frames provides expressing cell lines that consistently provide sufficient yields in faster time frames saving a long period. Furthermore, the expressing cell lines are produced with lower concentrations of selection and amplification agents than previously achievable. Accordingly lower levels of selection and amplification markers in the final expressing cells lines are observed.【課題】所望のタンパク質の発現細胞株を作製するのに必要な力価測定可能な選択圧のレベル、増幅サイクル数、およびそれにかかる時間を減らす方法を提供する。【解決手段】所望のタンパク質のオープンリーディングフレームのコドン最適化により、長期間を節約してより早い時間枠で、十分な収量を確実にもたらす発現細胞株を得る。さらに以前に達成可能なよりも低い濃度の選択・増幅薬剤で該発現細胞株を作製する。したがって、最終的な発現細胞株でより低いレベルの選択・増幅マーカーが観察される。【選択図】なし
