The present disclosure relates to anti-CD40 antibodies, such as humanized anti-CD40 antibodies, that can be used in various therapeutic, prophylactic and diagnostic methods. The antibody generally blocks the ability of CD40 to bind to CD154 and does so without activating CD40-expressing cells (eg, B cells). The antibodies or fragments thereof of the present invention can be used to reduce complications associated with organ or tissue transplantation.
