In certain embodiments methods of treating X-Linked agammaglobulinemia (XLA) in a mammal are provided where the methods comprise: i) providing differentiated T cells and/or stem/progenitor cells from the mammal； ii) performing a targeted insertion of a corrective BTK cDNA at the BTK gene locus in said cells to provide a corrected BTK gene in said cells； and iii) introducing said cells into said mammal where said corrected BTK gene is expressed in a physiologically regulated manner.L'invention concerne, dans certains modes de réalisation, des méthodes de traitement d'agammaglobulinémie liée à X (XLA) chez un mammifère, lesdites méthodes comprenant : i) la fourniture de cellules T différenciées et/ou de cellules souches/progénitrices provenant du mammifère； ii) la réalisation d'une insertion ciblée d'un ADNc de BTK correctif au niveau du locus de gène BTK dans lesdites cellules pour fournir un gène BTK corrigé dans lesdites cellules； et iii) l'introduction desdites cellules dans ledit mammifère, ledit gène BTK corrigé étant exprimé d'une manière physiologiquement régulée.
