A method of enhancing an immune response is provided comprising providing an immunogenic composition in combination with a CTLA-4 antagonist. Dendritic cell populations with reduced CTLA-4 expression are also provided. In a first embodiment, an immunogenic composition comprising at least a first antigen or antigen-sensitized dendritic cell and a CTLA-4 antagonist is provided. In a further embodiment, there is provided a method of inducing an immune response in a subject comprising administering to the subject an immunogenic composition in combination with a CTLA-4 antagonist.
