The present invention relates to nucleic acid molecules comprising a low repeat nucleotide sequence encoding a proline / alanine rich amino acid repeat sequence. The encoded polypeptide comprises a repetitive amino acid sequence that forms a random coil. The nucleic acid molecule comprising the low repeat nucleotide sequence may further comprise a nucleotide sequence encoding a bioactive or pharmacologically active protein. Furthermore, the present invention provides selection means and methods for identifying said nucleic acid molecule comprising said low repetitive nucleotide sequence. The invention also relates to methods for preparing said nucleic acid molecules. Also provided herein are methods for preparing a drug conjugate with the encoded polypeptide or the encoded polypeptide using the nucleic acid molecules provided herein. The drug conjugate may comprise a physiologically or pharmacologically active protein or small molecule drug.
