PROBLEM TO BE SOLVED: To provide a method for measuring and determining the root activity of a plant in order to quantitatively measure the activity of the plant root to instantaneously judge the growth state of the crop, by which the root activity can quickly, simply and quantitatively be measured only by bringing a cut plant root into contact with a CK reagent containing methylene blue. SOLUTION: The method for measuring and determining the activity of the plant root from the activity of a succinate dehydrogenase comprises immersing a constant amount of a cut plant root in a CK reagent comprising a sodium phosphate, a sodium citrate buffer solution as a respiratory buffer solution, sodium succinate as a respiratory substance, and extremely dilute methylene blue, and then comparing a change of color tone and a concentration of the color to measure the amount of released hydrogen.
