PROBLEM TO BE SOLVED: To provide a sterilization method usable for sterilization for a planting culture base of plants or seeds, and especially for mushroom cultivation, shortening time required to sterilize a culture base without lowering quality at the time of raising or harvesting mushroom, and to provide a sterilization device for the sterilization method. SOLUTION: This sterilization method comprises filling a culture base 3 in a culture bottle 1, inserting a hole-opening bar 5 having around jet orifices 7 of steam 15 into the inside of the culture base 3 and sterilizing the culture base 3 through jetting the steam 15 from the jet orifices 7 for a given time. The hole-opening bar 5 of a size wide enough to punch planting pits 8 of spawn is used so as to form the planting pits simultaneously with sterilizing. The sterilization device 9 is composed of a carrying part 10 to put the culture bottle 1 held at least in a container 11 thereon, and a hole-opening sterilizing part 12 set in the culture base 3 in the culture bottle 1 and punching the planting pits 8 of spawn simultaneously with sterilizing the culture base 3. The hole-opening bar 5 has an inlet 51 of the steam 15 on the upper side and a guide passage 6 of steam 15 in the inside.
