The present invention provides a monoclonal antibody that specifically binds human PD-1 with high affinity. Also provided is a nucleic acid molecule encoding an antibody of the invention, an expression vector, a host cell, and a method of expressing an antibody of the invention. The PD-1 antibodies of the present invention inhibit binding of PD-L1 to PD-1, thereby generally regulating the immune response and in particular the immune response mediated by TcR and CD28.
