A method of making triterpenoids from petri dish cultured Antrodia cinnamomea is disclosed, which is to extract triterpenoids from a extraction tank containing 1-2kg Antrodia cinnamomea under a pressure of 2,000-4,000 psi, a temperature of 40-60 DEG C. and a flow rate of 6-9 L/hr, to mix supercritical CO2 and ethanol with a volume ratio of 1:0.1-0.2 (v/v) to a saturated state; to send the Antrodia cinnamomea extract to a chromatographic column for separation, and to collect an active ingredient of the triterpenoid-rich compound at the top of the chromatographic column. Tests on bioactivity of the triterpenoids including cytotoxicity test, cell morphology analysis, cell cycle and apoptosis test, and cell motility have been conducted to verify the anti-cancer effect of the triterpenoids compound.本發明係提供一種製備皿培式牛樟芝中抗癌成分三萜類化合物之方法,係在壓力2000-4000psi、溫度40-60℃之操作條件下,將超臨界CO2流體與乙醇1:0.1-0.2(v/v)混合至飽和狀態,以流速6-9L/hr通入含1-2kg牛樟芝之萃取槽內進行萃取,萃取後再流入一層析管柱進行分離,可於層析管柱頂部得到高濃度三萜類化合物活性成分,並透過細胞毒性試驗、細胞型態變化的分析、細胞週期與細胞凋亡檢測、細胞爬行轉移等檢測,證實所製備之三萜類化合物具有抗癌效果。
