An antibody, or an antigen-binding fragment thereof, that binds to C5 of human complement for use in a method of treating paroxysmal nocturnal hemoglobinuria (PNH) or atypical hemolytic-uremic syndrome (aHUS) in a human, comprising the method: (i) administering to the human a siRNA specific for the mRNA encoding human C5 to thereby reduce the concentration of C5 in the human serum, followed by； (ii) administering the C5 antibody or fragment thereof to the human, wherein a reduced concentration of C5 in the human serum increases the serum half-life of the C5 antibody or fragment thereof administered to the human.Un anticuerpo, o un fragmento de unión a antígeno del mismo, que se une al C5 del complemento humano para su uso en un método para tratar hemoglobinuria paroxística nocturna (PNH) o síndrome hemolítico-urémico atípico (aHUS) en un ser humano, comprendiendo el método: (i) administrar al ser humano un ARNip específico para el ARNm que codifica C5 humano para reducir de ese modo la concentración de C5 en el suero del ser humano, seguido de； (ii) administrar al ser humano el anticuerpo de C5 o fragmento del mismo, en donde una concentración reducida de C5 en el suero del ser humano aumenta la semivida en suero del anticuerpo de C5 o fragmento del mismo administrado al ser humano.
