The method for screening transgenic animals generated by ultrasound image-guided gene delivery technique using in vivo imaging of reporter gene expression
PURPOSE: A method for selecting a transgenic animal using UIGD(ultrasound image-guided gene delivery) technique is provided to simply distinguish a target gene-delivered animal. CONSTITUTION: A method for selecting a transgenic animal in which target proteins are specifically expressed in brain cells comprises: a step of preparing a recombinant retrovirus vector containing one or more genes encoding target proteins and reporter gene； a step of preparing the retrovirus particles using vescular stomatitis virus(VSV-G) glycoprotein expression vector and recombinant retrovirus vector； a step of exposing the uterus by ultrasonic wave and performing microinjection of the retrovirus particles to embryonic ventricle； a step of developing the microinjected embryo； and a step of selecting an individual expressing a reporter gene in the embryo or adult body.본 발명은 초음파 영상 기반의 유전자 전달(Ultrasound Image-guided Gene Delivery, UIGD) 기술을 이용한 형질전환 동물의 선별 방법에 관한 것으로서, 구체적으로 초음파 영상을 기반으로 유전자를 전달함으로써 ED 8.5 내지 9.5일의 초기 발생 단계의 마우스 배아의 뇌실에 유전자를 주입할 수 있고, 마우스 백혈병 바이러스(Murine Leukemia Virus, MLV) 기반 레트로바이러스 벡터, 또는 이의 3'LTR이 마우스 줄기세포 바이러스(Murine Stem Cell Virus, MSCV)의 3'LTR로 치환된 레트로바이러스 벡터는 바이러스 역가가 향상되어 고농도로 생산 가능하며, MLV 또는 MSCV의 3'LTR을 사용하고 목적 단백질 및 리포터 유전자가 포함된 레트로바이러스 벡터와 함께, 수포성 구내염 바이러스-G(vescular stomatitis virus, VSV-G) 단백질을 외피 단백질로서 사용하여 제조한 레트로바이러스를 마우스 배아의 뇌실에 주입함으로써, 상기 목적 단백질이 초기 발생 단계의 마우스 배아 및 성체의 뇌세포에서 안정적으로 발현됨을 확인하였고, 한편, 목적 유전자를 포함하는 재조합 아데노바이러스를 마우스 배아의 뇌실에 주입하여, 상기 목적 단백질이 마우스 배아의 뇌세포에서 발현됨을 확인하였다. 이러한 유전자 전달 기술에 있어서 리포터 유전자를 함께 전달하여 그 발현을 확인함으로써 살아있는 상태에서도 목적 유전자가 전달된 형질전환 동물을 간편하게 구별하였으므로, 최적화된 UIGD 기술 및 리포터 유전자를 포함하는 레트로바이러스 또는 아데노바이러스 유래의 유전자 전달체는 목적 단백질이 뇌세포 특이적으로 발현하는 형질전환 동물의 선별에 유용하게 이용할 수 있다.
