METHOD TO SEPARATE AND PURIFY VEGETABLE ASCORBIC ACID-2-GLUCOSIDE FROM LYCIUM CHINENSE MILL EXTRACT BY UTILIZING THE CHARACTERISTICS OF SUGAR-BINDING PROTEIN
The present invention relates to a method for isolating and purifying plant ascorbic acid-2-glucoside from a Lycium chinense mill extract using characteristics of sugar-binding protein which purifies and isolates vitamin C derivative having higher purity than conventional methods by extracting the plant ascorbic acid-2-glucoside from various biologically active compounds contained in Lycium chinense mill fruits and applying endemic characteristics owned by the sugar-binding protein to an affinity chromatography. The present invention comprises: a first step of washing and grinding dried Lycium chinense mill fruits; a second step of extracting ascorbic acid derivative from the ground Lycium chinense mill fruits using an ethanol solvent; a third step of filtering the extract produced according to the second step and isolating the solvent; and a fourth step of concentrating the extract from which the solvent is isolated using a concentrator.본 발명은 구기자의 열매에 함유된 여러 가지 생리적 활성물질 중 식물성의 아스코르빈산-2-글루코사이드를 추출하되 당 결합 단백질이 갖고 있는 고유의 특이성을 친화성 크로마토그래피 방법에 적용시킴으로써 기존 방식보다 높은 순도의 비타민 C 유도체를 정제 및 분리하는 당 결합 단백질의 특성을 이용하여 구기자 추출물로부터 식물성 아스코르빈산-2-글루코사이드를 분리 및 정제하는 방법에 관한 것이다.
