PURPOSE: A nerve cell and a hair cell differentiated from amnion-derived mesenchymal stem cells are provided to treat hearing loss caused by damage of hair cells and to be used in regenerative medicine.CONSTITUTION: A method for differentiating amnion-derived mesenchymal stem cells into nerve cells and hair cells comprises a step of culturing the mesenchymal stem cell in a medium containing nerve growth factor AT 36-38 deg. C under the presence of 4-6% carbon dioxide for 5-15 days. The nerve growth factor is selected among GDNF(Grial-Derived Neurotrophin Factor), BDNF(Brain-Derived Neutrophin Factor), and neurotrophin-3(NT-3). The medium additionally contains 1-3 wt%(v/v) of B-27 supplement, 1-3 mM of L-glutamine, and 0.5-2 wt%(v/v) of penicillin-streptomycin. A composition for cell therapy for preventing or treating hearing loss contains the nerve cells and hair cells as active ingredients.COPYRIGHT KIPO 2013본 발명은 태반조직의 양막 유래 간엽줄기세포로부터 신경세포 및 유모세포를 분화시키는 방법에 관한 것으로서, 보다 구체적으로 본 발명은 양막 유래 간엽줄기세포를 신경성장인자가 포함된 배지에 배양하는 단계를 포함하는 양막 유래 간엽줄기세포로부터 신경세포와 유모세포를 분화시키는 방법 및 상기 분화된 신경세포 및 유모세포를 유효성분으로 포함하는 난청의 예방 또는 치료용 조성물에 관한 것이다. 본 발명에 따라 양막 유래 간엽줄기세포로부터 분화된 신경세포 및 유모세포는 유모세포의 손상으로 야기되는 난청을 치료하기 위한 세포 대체 요법 및 재생의학 분야에 사용될 수 있는 효과가 있으며 난청 치료를 위한 신약개발의 소재로도 사용할 수 있을 뿐만 아니라 난청 치료와 관련된 기초 연구 분야에서도 널리 사용될 수 있는 효과가 있다.
