1/1 summary “bordetella pertussis capsule tohama, methods to produce a strain of bordetella pertussis and bordetella pertussis antigens, antigen, use of an antigen, and acellular vaccine” are recombinant bordetella pertussis strains derived from the tohama precursor strain are provided. unpublished strains are obtained by homologous recombination using an allelic exchange vector pss4245, which allows replacement of bacterial chromosome sections without leaving accessory mutations. the coding segment of the pt subunit s1 is replaced to introduce two mutations that cause inactivation of pt's toxic activity. This strain may be further modified to express larger amounts of rpt and / or prn. a second copy of the ptx grouping of the five ptx structural genes of the ptx-ptl operon, with their promoter and ptl terminator, and containing the above mutations can be inserted elsewhere on the chromosome. Moreover, a second copy of the prn gene can be inserted into the chromosome.
