A method of generating an isolated population of cells comprising anti-third party cells having a central memory T-lymphocyte (Tcm) phenotype, the cells being tolerance-inducing cells and/or endowed with anti-disease activity, and capable of homing to the lymph nodes following transplantation is disclosed. The method comprising: (a) contacting peripheral blood mononuclear cells (PBMC) with a third party antigen or antigens in the presence of IL-21 so as to allow enrichment of antigen reactive cells； and (b) culturing the cells resulting from step (a) in the presence of IL-21, IL-15 and IL-7 in an antigen free environment so as to allow proliferation of cells comprising the central memory T-lymphocyte (Tcm) phenotype.セントラルメモリーＴリンパ球（Ｔｃｍ）表現型を有する抗第三者細胞を含む単離された細胞集団を作製する方法であって、前記細胞は寛容性誘導細胞であり、かつ／または、抗疾患活性を備え、かつ、リンパ節へのホーミングを移植後に行うことができ、前記方法が、（ａ）末梢血単核細胞（ＰＢＭＣ）をＩＬ－２１の存在下において第三者抗原（１つまたは複数）と接触させて、その結果、抗原反応性細胞の濃縮を可能にするようにすること、および、（ｂ）工程（ａ）から生じる細胞を、抗原非存在の環境で、ＩＬ－２１、ＩＬ－１５およびＩＬ－７の存在下において培養して、その結果、前記セントラルメモリーＴリンパ球（Ｔｃｍ）表現型を含む細胞の増殖を可能にするようにすることを含む方法。【選択図】なし
