A method is provided for producing a plasmacytoid dendritic cells (pDCs),wherein hematopoietic stem and progenitorcells (HSPCs) are provided and incubated in a first medium comprisingcytokines and growth factor whereby the HSPCs aredifferentiated into precursor-pDCs and then adding interferons (IFNs) to thefirst medium to obtain a second medium whereby saidprecursor-pDCs are differentiated into pDCs. Furthermore, a technique isprovided for producing genetically modified pDCs, by initiallygenetically modifying HSPCs using transfection methods, includingelectroporation, to deliver sgRNA and Cas9 protein Moreover, apharmaceutical formulation and a vaccine is provided which comprises pDC orgenetically modified pDCs obtained according to that method.
