The present invention provides purified, recombinantly produced bacterial lipoprotein signal peptidase (Lsp) enzyme, and an in vitro assay for monitoring Lsp catalytic activity. The present invention also provides screening methods for identifying novel antibiotics and their therapeutic applications for treating bacterial infections. Furthermore, the present invention provides specific Lsp-inhibiting compounds that can be used as bactericides in treating diseases resulting from bacterial infections.
