Methods and compositions for immunotherapy of inflammatory and immune-dysregulatory diseases, using multispecific antagonists that target at leasttwo differentmarkers are disclosed. When the multispecific antagonist reacts specificallywith a targetassociated with an inflammatory or immune-dysregulatory disorder, with apathologicangiogenesis or cancer, or with an infectious disease, it also bindsspecifically with at leastone proinflammatory effector of the immune system or at least one coagulationfactor. Thus,the multispecific antagonist contains at least one binding specificity relatedto the diseasedcell or condition being treated and at least one specificity to a component ofthe immunesystem. The multispecific antagonists are used in the treatment of variousdiseases that aregenerated or exacerbated by, or otherwise involve, proinflammatory effectorsof the innateimmune system or coagulation factors.
