The invention relates to improved compositions and methods for treating diseases, such as cancer, by providing a combination therapy comprising a TGFβ Receptor (TGFβR) antagonist comprising a domain antibody and a cell immunotherapy, in particular where the cell immunotherapy is an immunomodulatory cell expressing a chimeric antigen receptor (CAR) or a modified T cell receptor (TCR). The invention further relates to polynucleotides, expression vectors and immunomodulatory cells comprising the combination therapy, as well as methods of generating said immunomodulatory cells.
