The present invention relates to a polynucleotide encoding a modified neurotoxin polypeptide comprising a light chain and a heavy chain of a modified neurotoxin, wherein the modified light chain has at least one modification that confers alteration of cleavage by a calpain protease. Furthermore, the present invention includes vectors and host cells containing the polynucleotide of the present invention and the polypeptide encoded by the above polynucleotide.
