The present invention relates to porous PLGA micro-particles used for the sustained tiotropium release for treatment of chronic obstructive pulmonary diseases and a preparation method thereof. According to the present invention, the PLGA micro-particle preparation method includes the steps of: a) dissolving the tiotropium, ammonium bicarbonate (AB), and the biodegradable polysaccharide in water to prepare am internal aqueous phase b) applying a biodegradable polymer in an organic solvent to prepare an organic phase c) emulsifying the internal aqueous phase obtained in the step a) and the organic phase obtained in the step b) to prepare the water in oil emulsion (W/O) and d) injecting the W/O obtained in the step c) in an external aqueous phase including an aqueous coating material to obtain the water in oil in water (W/O/W) through multi-emulsion. The porous PLGA micro-particles used for the sustained tiotropium release have low density and large particle size mot to be removed by macrophages and remain in the lungs for an extended period of time.COPYRIGHT KIPO 2015본 발명은 만성 폐쇄성 폐질환 치료를 위한 티오트로피움의 서방형 방출을 위한 다공성 PLGA 미세입자 및 이의 제조방법에 관한 것이다. 본 발명의 다공성 PLGA 미세입자는, a) 티오트로피움, 중탄산 암모늄(ammonium bicarbonate AB) 및 생분해성 다당류를 물에 용해시켜 내부 수상을 제조하는 단계 b) 생분해성 고분자를 유기용매에 가하여 유기상을 제조하는 단계 c) 상기 단계 a)에서 얻어진 내부 수상과 단계 b)에서 얻어진 유기상을 유화하여 유중수적형(water in oil emulsion W/O )을 제조하는 단계 및 d) 상기 단계 c)에서 얻어진 유중수적형을 수성코팅물질이 포함된 외부수성상에 주입하여 복합유제(water in oil in water W/O/W) 형태로 다중유화 시키는 단계를 포함하여 이루어지는 것을 특징으로 한다. 본 발명의 티오트로피움의 서방형 방출을 위한 다공성 미세입자의 경우 낮은 밀도와 큰 입자크기를 갖는 다공성 미세입자를 제공하여 식세포로 인한 제거를 막고 폐 내부에 미세입자가 길게 잔존하는 효과를 가진다.
