#$%^&*AU2020102159A420201015.pdf#####Abstract Disclosed is a recombinant Newcastle disease virus heat-resistant vaccine strain expressing truncated Fiber 2 protein of Fowl Adenovirus serotype 4, a preparation method and application thereof which belongs to the field of vaccine genetic engineering. Reverse genetic manipulation technology of RNA virus is used to genetically modify the NDV LaSota strain at the transcription plasmid level. Firstly, an HN gene is replaced with an HN gene of a heat-resistant TSO9-C strain, and then a truncated Fiber2 gene (961-1440bp) of a Fowl Adenovirus serotype 4 is inserted between P and M genes thereof. The modified transcription plasmid is used to transfect host cells. A NDV rLS-tFib2-C strain is rescued and obtained. The vaccine strain has remarkable heat resistance properties, which thus can greatly reduce the dependence on the cold chain system, save storage and transportation costs and improve the thermal stability of the vaccine. The vaccine strain does not express the complete Fiber2 protein, only a part of region intercepted, which specifically enhances the immune protection effect of the vaccine. The NDV rLS-tFib2-C strain can be used for preparation of heat-resistant live vaccine against Angraradisease and Newcastle disease.
