The present invention relates to a novel crystalline tenofovir disproxil salt and a preparation method thereof. According to the present invention, the crystalline tenofovir disproxil orotate is an anhydride and is allowed to resolve pharmaceutical problems caused by physical and chemical properties of tenofovir disproxil fumarate, and has excellent physical and chemical properties to be pharmaceutically used. The crystalline tenofovir disproxil orotate of the present invention has the same optical stability as the tenofovir disproxil fumarate is highly stable in harsh conditions with no corrosion, and is not harmful to the human body leading to stability when manufactured, and useful of mass production with ease of handling.본 발명은 신규한 결정성 테노포비어 디소프록실 염 및 이의 제조방법에 관한 것이다.본 발명에 따른 결정성 테노포비어 디소프록실 오로테이트는 무수물이며, 테노포비어 디소프록실 푸마레이트의 물리화학적 특성의 문제로 야기되는 제제학적 단점을 해결하여 제제학적으로 용이하게 사용되기 위해 물리화학적으로 우수한 성질을 가진다. 즉, 본 발명의 결정성 테노포비어 디소프록실 오로테이트는 테노포비어 디소프록실 푸마레이트와 동등 이상의 광 안정성을 갖고, 특히 가혹조건에서 매우 높은 안정성을 가지고 있고, 부식성이 없으며 인체에 유해하지 않아 제조상 안전성이 있고 취급이 용이하여 대량 생산에 매우 유용하다.또한, 본 발명은 유기용매를 사용하는 방법과 유기용매를 전혀 사용하지 않는 환경 친화적인 제조방법으로서 산업적으로 실행 가능한 제조방법을 제공한다.또한 본 발명에 의해 제조된 테노포비어 디소프록실 오로테이트는 항바이러스 치료제로 사용될 수 있으며 다른 항-HIV 활성 성분인 약물과 복합 조성물로 항-HIV 치료제로 사용될 수 있는 조성물을 만든다. 이때 같이 사용될 수 있는 약물로는 엠트리시타빈(Emtricitabine), 에파비렌즈(Efavirenz), 릴피비렌즈(Rilpivirenz), 코비시스테트(Cobicistat), 엘비테그라비르(Elvitegravir), 랄테그라비르(Raltegravir), 리토나비르(Ritonavir), 로피나비르(Lopinavir), 다루나비르(Darunavir), 아타자나비르(Atazanavir) 등 또는 이들의 염으로 한가지 약물 내지 6가지 약물과 함께 만든 복합 조성물로 HIV 감염의 치료제로 사용 될 수 있다.
