Provided are a cellulose hydrolase and a gene thereof obtained by screening a cDNA genomic library constructed withOrpinomycessp. Y102. The gene is 1071 base pairs long and comprises an open reading frame (ORF) for producing a cellulose comprising 357 amino acids by translation. A transformed cell and a carrier carrying the gene are provided. The gene is transferred toE. coliby transformation, such thatE. colican acquire activity of decomposing CMC, beta-glucan, and xylan. The cellulose hydrolase is a multifunction cellulose and is capable of decomposing cellubiose and directly decomposing fiber into glucose.本發明提供自瘤胃真菌Orpinomycessp. Y102所建構之cDNA基因庫中經篩選所獲得一種纖維素水解酵素基因,此基因全長為1071個鹼基對,包含一個開放閱讀架構(ORF),可轉譯出含有357個胺基酸纖維分解酵素;同時亦提供包含該基因之載體及轉形細胞。此外,進一步將此基因轉型至大腸桿菌後,可使大腸桿菌產生分解CMC、beta-glucan與xylan的活性,為一多功能的纖維分解酵素,且同時具有分解纖維二糖的能力,可直接將纖維質分解成葡萄糖。
