This invention mainly discloses a queen bees breeding estimation method which is used to solve a problem for a conventional process of man-made breeding queen bees. The method comprises a preparing step, an estimating step, a selecting step and a matching step. The selecting step is selecting a father source and a mother source corresponded to a maximum value of a plurality of elected average values as an elected assembly by a computer system to execute the matching step after executing a process of out the old queen bee and in a new queen bee by a user. The matching step is monitoring volume of a beehive through an audio receiver, if the volume rising above a threshold and then reducing below the threshold to use the elected assembly as a breeding assembly and output it, else deleting the elected assembly to re-execute the selecting step until outputting the breeding assembly or without any elected assembly.
