The present invention relates to a composition for treating malignant mesothelioma including a polyphenol-based compound extracted from grapes and an anticancer agent of clofarabine as active ingredients. More specifically, the present invention relates to a composition for treating malignant mesothelioma containing resveratol, a polyphenol-based compound of grapes and clofarabine, an anticancer agent as active ingredients. It is found that complex administration of resveratrol which is a polyphenol-based compound and an anticancer agent of clofarabine maintains minimum toxicity in normal mesothelial cells of Met-5A while inducing strong synergistic apoptosis in malignant mesothelioma cells of MSTO-211H, and it is proved that the synergistic toxic mechanism is carried out by a phenomenon of inducing apoptosis by inhibiting the activation of Nrf2, Sp1 and Akt, inducing the activation of p53, and changing the transcriptional activation of a regulatory gene at a molecular level. Resveratrol and clofarabine inhibits survivals of various cells and targets for malignant alteration and induces apoptosis, and therefore, can be effective as a novel agent for treating malignant mesothelioma.본 발명은 포도에서 추출한 폴리페놀계 화합물과 항암제 클로파라빈을 유효성분으로 하는 악성 중피종 치료용 조성물에 관한 것으로서, 보다 상세하게는 포도의 폴리페놀계 화합물인 레스베라트롤과 항암제 클로파라빈을 유효성분으로 하는 악성 중피종 치료용 조성물에 관한 것이다.폴리페놀계 화합물 레스베라트롤과 항암제 클로파라빈의 복합 투여가 정상 중피세포 Met-5A에는 최소한의 독성을 유지하는 반면에 악성 중피종 세포 MSTO-211H에 강한 상승적 세포사를 유도하고 있음을 발견하였으며, 이러한 상승적 독성기전으로써 Nrf2, Sp1 및 Akt의 활성 억제와 p53의 활성화를 유도하고 조절유전자의 전사 활성을 변화시켜 세포자멸사를 유도함으로써 이루어 진다는 현상을 분자수준의 연구를 통해 증명하였다. 레스베라트롤과 클로파라빈은 다양한 세포생존 및 악성화 표적들을 억제하고, 세포자멸사를 유도시킴으로써 새로운 악성 중피종의 치료제로써 유용할 것이다.
